试验以苹果品种新红星、奥查金、美铃7为试材,对影响无菌繁殖系建立的两个主要因素即污染率和分化率进行了研究。结果表明:在4月下旬,各品种均已展叶4~5片时接种生长点,污染率较低,成活率较高,且易于生长分化。并发现不同品种间分化难易程度有很大差异,新红星是一个较难分化的品种,通过研究,其较适的组培条件是:改良C17+BA0.5mg/L+IBA0.2mg/L+CH300mg/L十蔗糖30g/L为起始培养基,接种后先光培养7d再暗培养7d,然后一直光培养直至分化。
In this experiment, the apple varieties of New Red Star, Ao Chajin, Meiling 7 were used as materials to study the two main factors that affect the establishment of asexual propagation line, namely, the pollution rate and the differentiation rate. The results showed that in late April, all the cultivars were inoculated with 4 to 5 leaves, with lower pollution rate, higher survival rate and easy growth and differentiation. The results showed that there was a great difference in the differentiation degree among different varieties. The new red star was a more difficult to differentiate. The suitable tissue culture conditions were as follows: modified C17 + BA0.5mg / L + IBA0.2mg / L + CH300mg / L decane 30g / L as the initial medium, after the first light-cultured 7d after inoculation and dark culture 7d, and then has been light until the differentiation.
