目的 探讨表皮生长因子受体 (EGFR)和 c- erb B- 2基因表达与人脑胶质瘤发生发展的关系 .方法 采用原位杂交方法检测 5 2例人脑胶质瘤、 2株人脑胶质瘤细胞系及 8例正常人脑组织中 EGFR和 c- erb B- 2基因 m RNA的表达 .结果 EGFR m RNA阳性率在高恶性度 ( ~ 级 )胶质瘤为 79.2 % ,低恶性度 ( ~ 级 )胶质瘤为 5 0 .0 % ,二者差异显著 (P<0 .0 5 ) ;c- erb B- 2 m RNA阳性率在高恶性度胶质瘤为 83.3% ,低恶性度胶质瘤为 71.4% ,二者无显著差别 (P>0 .0 5 ) ;EGFR和 c- erb B- 2 m RNA表达均随胶质瘤恶性程度增高而增强 (P<0 .0 1) .EGFR和 c- erb B- 2 m RNA在 2株人脑胶质瘤细胞系中均有表达 ,在 8例正常人脑组织中均无表达 . EGFR和 c- erb B- 2 m RNA表达结果密切相关 (P<0 .0 5 ) .结论 EGFR和 c- erb B- 2基因在人脑胶质瘤中普遍表达 ,且随胶质瘤恶性程度增高而表达增强 ,EGFR和 c- erb B- 2基因共表达在人脑胶质瘤发生发展过程中发挥重要作用
Objective To investigate the relationship between the expression of epidermal growth factor receptor (EGFR) and c-erb B-2 gene and the development of human glioma.Methods In situ hybridization was used to detect 52 human gliomas and 2 human brain Glioma cell lines and 8 normal human brain tissues.Results The positive rate of EGFR m RNA was 79.2% in glioma with high grade (grade), low malignancy (~ Grade) glioma was 50.0%, the difference was significant (P <0.05); the positive rate of c-erb B- 2 m RNA in the high-grade glioma was 83.3%, low There was no significant difference between the two groups (P> 0.05). The expressions of EGFR and c-erb B-2 mRNA both increased with the malignant degree of glioma (P <0. 0) 1) .EGFR and c-erb B-2 m RNA were expressed in both human glioma cell lines and none in 8 normal human brain tissues.Expression of EGFR and c-erb B- 2 m RNA (P <0.05) .Conclusion The expression of EGFR and c-erb B-2 gene is generally expressed in human glioma, which is increased with the malignant degree of glioma.The expression of EGFR and c-erb B-2 gene co-expression in human brain glue Development play an important role in tumor occurrence
