目的:建立莲威阿那奇处方水提浸膏中多糖的含量测定方法。方法:采用苯酚-硫酸显色,紫外-可见分光光度法测定莲威阿那奇处方水提浸膏中多糖的含量。结果:样品经苯酚-硫酸显色后在485 nm处有最大吸收,溶液的吸收度与葡萄糖含量在0.02~0.1 g.L-1呈良好线性关系,回归方程A=8.230C-0.016 3(r=0.999 8),平均回收率为99.07%,RSD0.99%,浸膏中多糖的平均含量为46.12%。结论:采用的苯酚-硫酸显色法操作简便,结果准确,稳定性和重现性好,可作为莲威阿那奇处方水提浸膏中多糖的含量测定方法。
Objective: To establish a method for the determination of polysaccharide in Lianwei Aniqi prescription water extract extract. Methods: The content of polysaccharide in Lianwei Aniqi prescription water extract was determined by phenol - sulfuric acid colorimetry and UV - visible spectrophotometry. Results: The sample showed maximum absorption at 485 nm after phenol-sulfuric acid colorization. The absorbance of the sample showed a good linear relationship with the glucose content of 0.02-0.1 gL-1. The regression equation was A = 8.230C-0.016 3 (r = 0.999 8), the average recovery was 99.07%, RSD0.99%, the average content of polysaccharides in extract was 46.12%. Conclusion: The phenol-sulfuric acid colorimetric method is simple, accurate, stable and reproducible. It can be used as a method for the determination of polysaccharides in Lianwei Aniqi prescription water extract.
