目的探讨牛蒡子苷对糖尿病肾病(DN)大鼠肾小球滤过屏障中nephrin、podocin和乙酰肝素酶(HPSE)表达的影响。方法将SD大鼠制备成DN实验模型,随机分为模型组、牛蒡子苷小、大剂量组(ig 40、60 mg.kg-1.d-1)正常对照组。8周后观察各组血糖、24 h尿蛋白定量及肾脏指数,光镜下观察肾组织的病理改变,采用免疫组化和Real time-PCR检测肾皮质中nephrin、podocin、HPSE蛋白和mRNA的表达。结果牛蒡子苷可降低DN大鼠尿蛋白的排泄,降低肾脏指数,改善肾脏病理,使nephrin、podocin蛋白和mRNA的表达增加,HPSE蛋白的表达降低。结论牛蒡子苷对DN大鼠具有一定的肾脏保护作用,其作用机制可能与调控肾小球滤过屏障中相关蛋白的表达有关。
Objective To investigate the effects of arctiin on the expression of nephrin, podocin and heparanase (HPSE) in the glomerular filtration barrier of diabetic nephropathy rats. Methods SD rats were made into experimental model of DN and randomly divided into model group, small and high dose group of arctiin (ig 40,60 mg.kg-1.d-1) normal control group. After 8 weeks, the levels of blood glucose, 24 h urinary protein and kidney index were observed. The pathological changes of renal tissues were observed under light microscope. The expressions of nephrin, podocin and HPSE protein and mRNA in renal cortex were detected by immunohistochemistry and Real time-PCR . Results Arctiin could reduce excretion of urinary protein, reduce renal index and improve renal pathology in DN rats, increase the expressions of nephrin and podocin protein and mRNA, and decrease the expression of HPSE protein. Conclusion Arctiin has a protective effect on kidney of DN rats, and its mechanism may be related to the regulation of the expression of related proteins in the glomerular filtration barrier.
