目的 :通过研究异硫氰酸萘酯 (ANIT) )所致的急性肝内胆汁淤积性肝损伤兔模型血清和胆汁中TNF a、IL 6的代谢变化 ,来进一步探讨微生态制剂 乐托尔对该模型的作用机制。方法 :按 2 0 0mg/kg一次性灌服ANIT来诱发新生白兔急性肝内胆汁淤积性肝损伤病变 ,分别检测中毒后 2 4、4 8h及用乐托尔散剂治疗后血清和胆汁中总胆红素 (TB)、丙氨酸转氨酶 (ALT)、肿瘤坏死因子 a(TNF a)、白细胞介素 6(IL 6 )的值。结果 :在乐托尔组中发现 ,灌服ANIT 4 8h后血清和胆汁中的TNF a、IL 6、ALT、TB值均较中毒 4 8h组明显降低 (P值均小于 0 0 5 ) ;各组胆汁中TNF a、IL 6的浓度均明显高于血清中浓度。结论 :微生态制剂能够改善肝功能 ,增加胆汁的排泄量 ,使炎性细胞因子的产生量显著减少。
OBJECTIVE: To investigate the metabolic changes of TNFa and IL-6 in sera and bile of rabbits with acute intrahepatic cholestatic liver injury induced by naphthalene isothiocyanate (ANIT) The mechanism of action of this model. Methods: One-time ANOVA at 200 mg / kg was used to induce acute intrahepatic cholestatic liver injury in neonatal rabbits. The levels of total serum and bile after 24 and 48 hours of poisoning Bilirubin (TB), alanine aminotransferase (ALT), tumor necrosis factor a (TNFa), and interleukin 6 (IL 6). Results: In the Le Tolle group, TNFα, IL 6, ALT and TB in serum and bile were significantly decreased after 48 hours of ANIT treatment (P <0.05) Group bile TNF a, IL 6 concentrations were significantly higher than the serum concentration. Conclusion: The probiotics can improve liver function, increase the excretion of bile and decrease the production of inflammatory cytokines.
