目的:建立测定克淋通胶囊中槲皮素、盐酸小檗碱和盐酸巴马汀含量的高效液相色谱法。方法:采用高效液相色谱法,Agilent Eclipse XDB-C18色谱柱(4.6 mm×250 mm,5μm);流动相:乙腈-0.033 mol·L-1磷酸二氢钾溶液(每100 ml加十二烷基硫酸钠0.1 g)(50∶50);流速:1.0ml·min-1;紫外检测波长:345 nm。结果:槲皮素、盐酸小檗碱和盐酸巴马汀线性范围分别为0.020 44~0.306 6μg、0.042 97~0.644 5μg、0.020 10~0.301 5μg,线性关系良好(依次r=0.999 6,0.999 7,0.999 5),平均回收率分别为97.41%,98.72%,97.30%(n=9),RSD分别为0.5%,0.9%,0.6%。结论:该方法准确可靠,可重复性好,为克淋通胶囊的质量控制提供了方法依据。
Objective: To establish a HPLC method for the determination of quercetin, berberine hydrochloride and palmatine hydrochloride in Kailuotong Capsules. METHODS: High performance liquid chromatography (HPLC) was performed on an Agilent Eclipse XDB-C18 column (4.6 mm × 250 mm, 5 μm). The mobile phase consisted of acetonitrile-0.033 mol·L -1 potassium dihydrogen phosphate solution Sodium Sulfate 0.1 g) (50:50); Flow rate: 1.0 ml · min-1; UV detection wavelength: 345 nm. Results: The linear ranges of quercetin, berberine hydrochloride and palmatine hydrochloride were 0.020 44 ~ 0.306 6μg, 0.042 97 ~ 0.644 5μg and 0.020 10 ~ 0.301 5μg, respectively, with good linearity (r = 0.999 6,0.999 7, 0.999 5). The average recoveries were 97.41%, 98.72% and 97.30% (n = 9), respectively. The RSDs were 0.5%, 0.9% and 0.6% respectively. Conclusion: The method is accurate and reliable with good repeatability and provides a method basis for the quality control of Kelixong capsule.
