目的观察脉冲电场对荷黑素瘤小鼠肿瘤生长的抑制作用及对心肌细胞和心脏电活动的影响。方法皮下移植B16黑素瘤细胞建立荷黑素瘤小鼠模型,随机分为4组。治疗组分别采用1 ms、3 ms、5 ms脉冲宽度,100 V/cm场强、10 Hz频率的脉冲电场,10 min/d连续刺激15 d。记录小鼠的心电活动,游标卡尺测定肿瘤体积,HE染色观察肿瘤病变情况,采用实时荧光定量PCR检测肿瘤组织中增殖细胞核抗原(PCNA)mRNA的水平,免疫荧光组织化学技术检测肿瘤组织PCNA蛋白的表达,透射电子显微镜观察心脏组织的超微结构改变,ELISA测定小鼠血清中肌钙蛋白T(c Tn T)和肌酸激酶同工酶(CK-MB)的水平。结果与对照组相比,电刺激第7天后,各治疗组肿瘤体积均减小;治疗组瘤细胞胞质内充满黑素颗粒;各治疗组PCNA mRNA和蛋白表达均降低,其表达强度变化随着脉冲宽度增加而增强;3 ms组、5 ms组小鼠心肌细胞出现损伤,且血清中c Tn T、CK-MB表达均明显高于对照组。结论脉冲电场可抑制荷黑素瘤小鼠的肿瘤生长,脉冲宽度增加,会加重心肌细胞的损伤。
Objective To observe the inhibitory effect of pulsed electric field on tumor growth in mice bearing melanoma and its effect on cardiomyocyte and cardiac electrical activity. Methods B16 melanoma cells were transplanted subcutaneously to establish a mouse model of melanoma and were randomly divided into 4 groups. The treatment group were pulsed electric fields of 1 ms, 3 ms, 5 ms, 100 V / cm field and 10 Hz respectively for 10 days. The electrocardiogram (ECG) activity of the mice was recorded. The tumor volume was measured by vernier caliper, the pathological changes of the tumor were observed by HE staining. The level of proliferating cell nuclear antigen (PCNA) mRNA in the tumor tissue was detected by real-time fluorescence quantitative PCR. The ultrastructural changes of cardiac tissue were observed by transmission electron microscopy. The levels of cTnT and CK-MB in serum were determined by ELISA. Results Compared with the control group, after 7 days of electrical stimulation, the tumor volume of each treatment group decreased; the cytoplasm of treatment group was filled with melanin granules; the expression of PCNA mRNA and protein in each treatment group decreased, The pulse width increased and enhanced; 3 ms group, 5 ms group of myocardial cell injury, and serum cTn T, CK-MB expression were significantly higher than the control group. Conclusion Pulsed electric field can inhibit tumor growth in mice bearing melanoma. Increasing the pulse width will aggravate the injury of cardiac myocytes.
