目的 :研究化疗对非小细胞肺癌PCNA表达和肿瘤细胞DNA含量的影响 ,及PCNA阳性指数与DNA含量的关系。方法 :用免疫组化S -P法检测两配对组各 18例非小细胞肺癌PCNA表达 ,并用图像分析系统进行肿瘤DNA分析、倍体测量。结果 :化疗后组与对照组肿瘤在PCNA阳性指数 (PI)、DNA指数、平均DNA质量、S期比率及 >5C细胞百分比均有显著性差异 (P <0 .0 5,P <0 .0 1)。且PI与DNA含量之间呈正相关关系。但两组间异倍体肿瘤率无显著差异 (P >0 .0 5)。结论 :联合化疗影响肿瘤细胞PCNA表达及DNA含量 ,即降低了肿瘤的增殖活性
Objective: To study the effect of chemotherapy on PCNA expression and DNA content of tumor cells in non-small cell lung cancer, and the relationship between PCNA positive index and DNA content. METHODS: The expression of PCNA in 18 cases of non-small cell lung cancer was detected by immunohistochemical S-P method. The DNA analysis and ploidy measurements were performed using an image analysis system. Results: There were significant differences in PCNA positive index (PI), DNA index, average DNA quality, S-phase ratio, and percentage of >5C cells between the chemotherapy group and the control group (P < 0.05, P <0. 1). There was a positive correlation between PI and DNA content. However, there was no significant difference in the aneuploid tumor rate between the two groups (P > 0.05). Conclusion : Combination chemotherapy can affect the expression of PCNA and DNA content in tumor cells, that is, it reduces the proliferation activity of tumors.
