目的:研究菝葜对便秘型肠易激综合征大鼠的干预作用及作用机理。方法:用冰水灌胃法复制大鼠便秘型肠易激综合征模型,将造模成功大鼠分为模型组,菝葜提取物高、中、低剂量组(浓度依次为80g/kg、40g/kg、20g/kg,按生药量计),阳性对照组,另设空白对照组。灌胃给药3周后,硝酸还原酶法测定大鼠血清NO含量变化,酶免法测定大鼠血浆CGRP含量变化,观察对大鼠结肠VIP、iNOS免疫组化染色的影响。结果:与模型组相比,菝葜提取物20g/kg(按生药量计)能够显著降低模型大鼠血清NO和血浆CGRP含量(P<0.05);显著升高模型大鼠结肠VIP免疫组化染色的平均灰度值(P<0.05);极显著升高模型大鼠结肠iNOS免疫组化染色平均灰度值(P<0.01)。结论:菝葜对便秘型肠易激综合征模型大鼠有明显的干预作用,其作用机理可能是通过调整模型大鼠血清NO、血浆CGRP、结肠VIP、iNOS等方面来实现的。
Objective: To study the intervention effect and mechanism of Cuscuta on constipation-induced irritable bowel syndrome in rats. Methods: The rats with constipation-induced irritable bowel syndrome model were duplicated with ice-water gavage. The successful model rats were divided into model group, high, medium and low dose groups of extract (with the order of 80g / kg, 40g / kg, 20g / kg, according to the crude drug amount), the positive control group, another set of blank control group. After 3 weeks of intragastric administration, the content of NO in serum was measured by nitrate reductase method. The content of CGRP in plasma of rats was measured by enzyme immunoassay. The effect of immunohistochemical staining on VIP and iNOS in colon was observed. Results: Compared with model group, 20g / kg of extract of Scutellaria baicalensis Georgi could significantly reduce the level of serum NO and plasma CGRP (P <0.05), and significantly increase the expression of VIP in colon of model rats (P <0.05). The average gray value of iNOS immunohistochemical staining in model group was significantly increased (P <0.01). Conclusion: Emu owl has a significant intervention effect on constipation-induced irritable bowel syndrome rats, and its mechanism may be achieved by adjusting the serum NO, plasma CGRP, VIP, iNOS and other aspects of the model rats.
