目的研究半边旗二萜类化合物5F(11α-羟基-15-氧-16-烯-对映贝克杉烷-19酸)对人高转移卵巢癌细胞系HO-8910PM细胞内核转录因子κBP65亚基[NF-κB(P65)]、Smad3蛋白及NF-κB(P65)、ETS-1mRNA表达的影响,探讨其抗肿瘤的作用机制。方法MTT法检测5F对HO-8910PM细胞增殖的影响;Westernblot分析NF-κB(P65)及Smad3蛋白的表达;RT-PCR分析NF-κB(P65)及ETS-1mRNA的表达。结果5F能抑制HO-8910PM细胞的增殖,抑制率随剂量的增加而增加,具有剂量依赖性;25~100μmol/L5F作用HO-8910PM细胞24h后,NF-κB(P65)、Smad3蛋白表达水平明显下降,并呈剂量效应关系;5F明显下调NF-κB(P65)mRNA的表达,轻微下调ETS-1mRNA的表达。结论5F抗肿瘤活性与NF-κB(P65)、Smad3蛋白和NF-κB(P65)、ETS-1mRNA的表达有关。
Objective To study the effects of diterpene 5F (11α-hydroxy-15-oxo-16-ene-p-terbulular-19 acid) on the expression of nuclear transcription factor κBP65 subunit of human highly metastatic ovarian cancer cell line HO-8910PM [ (P65)], Smad3 protein, NF-κB (P65), and ETS-1 mRNA expression, and explore its anti-tumor mechanism. Methods The effect of 5F on the proliferation of HO-8910PM cells was detected by MTT assay. The expressions of NF-κB (P65) and Smad3 protein were detected by Western blotting. The expressions of NF-κB (P65) and ETS-1 mRNA were detected by RT- Results 5F inhibited the proliferation of HO-8910PM cells in a dose-dependent manner. The expression of NF-κB (P65) and Smad3 in HO-8910PM cells was significantly increased after 25 ~ 100μmol / L 5F treatment 5F decreased the expression of NF-κB (P65) mRNA and down-regulated the expression of ETS-1 mRNA in a dose-dependent manner. Conclusion The antitumor activity of 5F is related to the expression of NF-κB (P65), Smad3 protein, NF-κB (P65) and ETS-1 mRNA.
