淫羊藿甙(icariin,ICA)是从小蘖科草本植物朝鲜淫羊藿中提取的主要活性成份,具有多种生物学效应,是一种新型的生物反应调节剂和诱导分化剂.但有关淫羊藿甙抗肿瘤作用的研究国外未见报道,而国内的报道仅限于白血病细胞.为进一步探讨ICA对肿瘤细胞恶性表型的逆转作用及其调控机制,我们将ICA作用于人高转移肺癌细胞PC,采用MTT、放免、流式细胞术、粘附实验、运动侵袭实验等多种手段进行了检测.结果表明,ICA虽直接抑瘤作用较弱,但能影响PG细胞周期的时相分布,阻止其由GO/G1期向S期移行,与对照组相比,ICA 200μg/ml处理24 h后,GO/G1期细胞增多,而S期细胞明显减少,由57.43%降低为33.07%.与此细胞周期变化的时相相吻合,细胞内cAMP水平升高(0.05>P>0.01),cGMP水平降低(P<0.01),cAMP/cGMP比值明显升高(P<0.01).根据最近的研究
Epimedium (icariin, ICA) is the main active ingredient extracted from the herb family Epimedium kombucha, and has a variety of biological effects. It is a novel biological response modifier and differentiation-inducing agent. The anti-tumor effect of L. edodes has not been reported abroad, but domestic reports are limited to leukemia cells. To further investigate the reversal effect of ICA on the malignant phenotype of tumor cells and its regulatory mechanisms, we applied ICA to human high-metastatic lung cancer cells. PC, using MTT, radioimmunoassay, flow cytometry, adhesion assays, exercise invasion assays and other means were tested. The results show that although ICA has a weak direct inhibitory effect, it can affect the phase distribution of PG cell cycle. To prevent its transition from GO/G1 phase to S phase, compared with the control group, after 24 h treatment of ICA 200 μg/ml, the number of GO/G1 phase cells increased, while S phase cells decreased significantly from 57.43% to 33.07%. This phase change of the cell cycle was consistent with the increase of intracellular cAMP levels (0.05>P>0.01), decreased cGMP levels (P<0.01), and significantly increased cAMP/cGMP ratio (P<0.01).
