目的:骨形成蛋白4(bonemorphogeneticprotein4,BMP4)体外诱导成年大鼠骨髓间充质干细胞分化为神经元的能力。方法:采用BMP4体外定向诱导第五代骨髓间充质干细胞,免疫组化鉴定神经丝蛋白(NF)、胶质纤维酸性蛋白(GFAP)的表达。结果:成年大鼠骨髓间充质干细胞受BMP4诱导后出现神经元样细胞,细胞免疫组化显示诱导出的神经元样细胞NF表达阳性,诱导后5h,12h,1d,3d,5d和7d的NF神经样细胞阳性率分别为(40±7)%,(71±6)%,(58±3)%,(53±6)%,(37±5)%,(13±2)%,诱导后12h,NF神经元样细胞阳性表达到高峰,与诱导后5h比较,差别有显著性(t=1.380~2.621,P<0.05)。结论:BMP4可以在体外诱导成年大鼠骨髓间充质分化为神经元。
OBJECTIVE: Bone morphogenetic protein 4 (BMP4) induces the differentiation of adult rat bone marrow mesenchymal stem cells into neurons in vitro. Methods: The fifth generation of bone marrow mesenchymal stem cells were induced by BMP4 in vitro and the expression of neurofilament protein (NF) and glial fibrillary acidic protein (GFAP) were identified by immunohistochemistry. Results: Bone marrow-derived mesenchymal stem cells of adult rats showed neuron-like cells after induced by BMP4, and the expression of NF was positive in neuron-like cells induced by BMP-4. After 5h, 12h, 1d, 3d, 5d and 7d The positive rates of NF-like neurons were (40 ± 7)%, 71 ± 6%, 58 ± 3%, 53 ± 6%, 37 ± 5%, 13 ± 2% At 12 h after induction, NF neuron-like cells peaked at the peak, which was significantly different from that at 5 h after induction (t = 1.380-2.621, P <0.05). Conclusion: BMP4 can induce bone marrow mesenchymal differentiation into neurons in adult rats.
