本实验的目的是评价血小板不同密度亚群对不同的血小板聚集剂的反应。研究结果表明,以间断梯度离心法,可将家兔血小板分为高密度(HDP)、中密度(IDP)和低密度(LDP)三个亚群。HDP亚群对胶原(COllgen)、花生四烯酸(AA)、血栓素(TXA2)同系物(STA2)、腺苷二磷酸(ADP)及Calciumionophore(A23178)的血小板聚集和ATP释放反应,都明显强于其它两个亚群(P<0.05或P<0.01)。单一血小板细胞内游离钙的测定结果也证实HDP亚群的细胞内钙动员显著高于IDP和LDP两亚群。结论,HDP亚群血小板的功能反应性强,对聚集剂最敏感。
The purpose of this experiment was to evaluate the response of different density platelets to different platelet aggregates. The results show that the rabbit platelets can be divided into three subgroups: high density (HDP), medium density (IDP) and low density (LDP) by means of discontinuous gradient centrifugation. The platelet aggregation and ATP release responses of HDP subpopulations to COllgen, arachidonic acid (AA), TXA2 homologues (STA2), adenosine diphosphate (ADP) and Calcium ionophore (A23178) Stronger than the other two subgroups (P <0.05 or P <0.01). The determination of intracellular free calcium in a single platelet also confirmed that the intracellular calcium mobilization of HDP subpopulations was significantly higher than both subpopulations of IDP and LDP. Conclusions: HDP subgroup platelets are highly functional and most sensitive to aggregating agents.
