目的:通过正交实验探讨治疗糖尿病地知复方的配伍规律。方法:采用一次性腹腔注射链脲佐菌素(STZ)造成糖尿病小鼠模型。选用L_9(3~4)正交表,进行实验分组。测定给药前后血糖,血清及肾组织的一氧化氮(NO)、丙二醛(MDA)含量。结果:根据正交实验结果分析得出,各味药降血糖作用大小依次为地黄→葛根→黄芪→知母,适宜的剂量水平为地黄_3葛_2黄芪_3知母_3;降低血清NO水平作用强弱顺序为知母→黄芪→葛根→地黄,适宜剂量水平为知母_3黄芪_3葛根_3地黄_2;降低肾组织NO作用强度依次为地黄→知母→黄芪→葛根,适宜剂量水平为地黄_2知母_3黄芪_1葛根_2。降低血清MDA含量作用强弱为黄芪→葛根→地黄→知母,最佳剂量水平为黄芪_3葛根_2地黄_2知母_1;降低肾组织MDA作用强弱顺序为知母/地黄→黄芪→葛根,适宜的剂量水平为知母_3地黄_2黄芪_3葛根_3。结论:对氧化损伤的改善,综合起来主要是知母和地黄起作用。当把血糖和氧化应激指标综合考虑时,可以得出,在此方中药味地黄起主要作用,最佳剂量水平为地黄_3知母_3黄芪_3葛根_3。
OBJECTIVE: To investigate the compatibility of diabetes mellitus and diarrhea by orthogonal experiment. Methods: A single intraperitoneal injection of streptozotocin (STZ) caused diabetic mice model. Select L_9 (3 ~ 4) orthogonal table, the experimental grouping. Blood glucose, serum and renal tissue were measured before and after administration of nitric oxide (NO), malondialdehyde (MDA) content. Astragalus → Pueraria → Radix Rehmanniae to Anemarrhena astigmatism, the best dose level was Astragalus_3 Pueraria root _2 Rehmannia_2 Anemarrhena_1; the order of decreasing the action of MDA in renal tissue was Anemarrhena / Rehmanniae; Astragalus → Pueraria, the appropriate dose level for the Anemarrhena _3 Rehmannia _3 Astragalus _3 Pueraria _3. Conclusion: The improvement of oxidative damage is mainly caused by the effects of Anemarrhena asphodeloides and Radix Rehmanniae. When the indicators of blood glucose and oxidative stress are taken into account, we can conclude that this side of the Chinese medicine taste yellow play a major role, the best dose level Rehmannia _3 Anemarrhena _3 Astragalus _3 Pueraria _3.
