目的:了解2008年韶关市B型流感病毒的流行及基因变异情况。方法:收集2008年韶关市流感监测及疫情资料,选取流感疫情中分离的B型毒株,提取病毒RNA,RT-PCR扩增病毒HA1基因,纯化产物进行核苷酸序列测定,使用DNA Start MegAlign等生物软件包进行序列分析。结果:2008年流感监测与疫情中一共分离到39株B型流感毒株,分离率为7.39%(39/528),与B型流感病毒相关的疫情达到当年疫情数量的一半(7/14),有4起完全由B型流感病毒引起,有3起同时检出B型流感病毒与A型流感病毒,爆发时间多集中在3月;B/SG/y010/2008与B/Shanghai/361/2002相比,3个氨基酸替换发生在抗原决定簇,与韶关以往毒株B/SG/173/1999比较4个氨基酸替换发生在抗原决定簇,均在196增加了一个糖基化位点。结论:B型流感病毒是韶关市2008年流感流行优势毒株,B/SG/y010/2008与B/Shanghai/361/2002相比发生了一定的变异,而与韶关以往毒株B/SG/173/1999相比则发生了抗原性漂移。
Objective: To understand the prevalence and genetic variation of influenza B virus in Shaoguan City in 2008. Methods: Influenza surveillance and epidemic situation data of Shaoguan City in 2008 were collected. Type B strain isolated from influenza epidemic situation was selected to extract viral RNA. The viral HA1 gene was amplified by RT-PCR and the product was purified for nucleotide sequence determination. DNA Start MegAlign Other biological software packages for sequence analysis. Results: A total of 39 influenza B strains were isolated from the influenza surveillance and epidemic outbreaks in 2008 with a separation rate of 7.39% (39/528). The outbreaks associated with influenza B virus reached half of the current year (7/14) , 4 were completely caused by influenza B virus, 3 were detected at the same time, influenza B virus and influenza A virus, the outbreak time and more concentrated in March; B / SG / y010 / 2008 and B / Shanghai / 361 / Compared with 2002, three amino acid substitutions occurred in the antigenic determinant. Compared with the previous strain B / SG / 173/1999 in Shaoguan, the four amino acid substitutions occurred in the antigenic determinant and both increased a glycosylation site at 196. Conclusion: Influenza B virus is the predominant strain of influenza in Shaoguan City in 2008, B / SG / y010 / 2008 and B / Shanghai / 361 / 173/1999 compared to the occurrence of antigenic drift.
