目的:于NB4细胞株中,对β-榄香烯衍生物ET(β-榄香烯13-色氨酸)进行抗肿瘤作用研究。方法:体外细胞培养,凋亡检测、流式细胞术和蛋白免疫印迹等技术进行机制研究。结果:ET在低于40μmol.L-1时可以抑制白血病细胞NB4生长并诱导凋亡;ET可以引起细胞内H2O2的蓄积并活化caspase-3;过氧化氢酶可以阻断ET引起的H2O2蓄积和凋亡。结论:ET主要通过HO的蓄积引起细胞凋亡。
OBJECTIVE: To study the antitumor effect of β-elemene derivative ET (β-elemene 13-tryptophan) in NB4 cell line. Methods: In vitro cell culture, apoptosis detection, flow cytometry and Western blotting were used to study the mechanism. Results ET inhibited the growth of NB4 cells and induced apoptosis when the concentration of ET was less than 40 μmol·L-1. ET induced the accumulation of H2O2 and activated caspase-3. Catalase could block the accumulation of H2O2 induced by ET and Apoptosis. Conclusion: ET mainly induces apoptosis through the accumulation of HO.
