目的探讨生物酶制剂JA治疗小鼠乳腺癌的可行性和安全性,为临床应用提供依据。方法噻唑蓝(MTT)法检测不同浓度JA制剂对MA782肿瘤细胞的作用,电镜观察JA制剂作用后肿瘤细胞的超微结构;用MA782细胞建立小鼠乳腺癌动物模型,共60只,随机分成3组(G1 ~G3),分别行瘤体内注射生理盐水、无水乙醇、制剂JA各0.2 ml,观察用药后肿瘤大小变化,于用药第1、第10天每组各处死8只小鼠,采血,检查肝肾功能,取肿块病检。结果 1/40倍浓度以上 JA制剂作用30 min即可将90%以上肿瘤细胞杀死;肿瘤消融效果G3>G2>G1,G2组出现ALT、 AST持续升高,G3组ALT、AST仅表现为一过性升高,其他各组间各指标差异无统计学意义。结论制剂JA对实验性小鼠乳腺癌(MA782肿瘤)有独特的消融效果,明显优于无水乙醇,是一种安全有效的生物酶消融制剂,具有潜在的临床应用前景。
Objective To investigate the feasibility and safety of biological enzyme preparation JA in the treatment of breast cancer in mice and provide the basis for its clinical application. Methods MTT assay was used to determine the effect of different concentrations of JA on MA782 tumor cells. The ultrastructure of tumor cells was observed by electron microscopy after the administration of JA. The mouse model of breast cancer was established by MA782 cells. A total of 60 mice were randomly divided into 3 groups Groups (G1-G3) were injected with normal saline, ethanol, respectively, the preparation of JA each 0.2 ml, observed tumor size changes after treatment, in the first day of treatment 10 mice were sacrificed in each group of 8 , Blood, liver and kidney function check, take mass disease seized. RESULTS: More than 90% of the tumor cells were killed by JA preparations at a concentration of 1/40 times. Tumor ablation effect was G3> G2> G1. ALT and AST were continuously increased in G2 group. ALT and AST in G3 group were only After transient increase, there was no significant difference among the other groups. Conclusion The preparation JA has a unique ablation effect on experimental mouse breast cancer (MA782 tumor), which is obviously better than absolute ethanol. It is a safe and effective biological enzyme ablation preparation and has potential clinical application prospects.
