目的:观察栀子对兔膝骨关节炎软骨组织中凋亡因子Bcl-2/Bax表达的影响。方法:50只大耳白兔随机分为5组,对照组、模型组、低剂量组、中剂量组、高剂量组,每组10只,除对照组外,各组用Hulth法制备骨关节炎模型,观察栀子干预后各组动物关节活动度、软骨大体形态及免疫组化法检测软骨组织中Bcl-2/Bax蛋白的表达情况。结果:各给药组关节活动度明显增大,与模型组比较差异有统计学意义(P<0.05);软骨组织中Bax下调,Bcl-2上调,与模型组比较有显著统计学意义(P<0.01)。结论:栀子有改善骨关节炎关节活动度的作用,上调软骨组织中Bcl-2蛋白的表达而下调Bax蛋白的表达,可能是抑制软骨细胞凋亡的机制之一。
Objective: To observe the effect of gardenia on the expression of apoptosis factor Bcl-2/Bax in rabbit knee osteoarthritis cartilage tissue. METHODS: Fifty rabbits were randomly divided into five groups: control group, model group, low-dose group, middle-dose group, and high-dose group. Ten rats in each group were used. In addition to the control group, the bone joints were prepared by Hulth method. The inflammation model was used to observe the degree of joint motion, gross cartilage morphology, and immunohistochemical staining of the Bcl-2/Bax protein expression in cartilage tissue after intervention with gardenia. RESULTS: The degree of joint motion in each group was significantly increased, and there was a statistically significant difference compared with the model group (P<0.05); Bax was down-regulated in cartilage and Bcl-2 was up-regulated, which was significantly statistically significant compared with the model group (P). <0.01). Conclusion: Gardenia jasminoides can improve the degree of osteoarthritis joint activity. Up-regulating the expression of Bcl-2 protein in cartilage tissue and down-regulating the expression of Bax protein may be one of the mechanisms that inhibit the apoptosis of cartilage cells.
