目的:在体外实验条件下观察氨对星状胶质细胞超微结构的影响。材料与方法:用NH_4CI造成高NH_4+环境,对体外培养之星形胶质细胞的形态变化进行了超微结构观察。结果:高NH_4+环境下,星形胶质细胞首先表现出损伤性变化:细胞电子密度降低,线粒体、内质网等细胞器肿胀,在胞质内形成许多单位膜包绕的电子密度降低的空泡区域。以后随着氨浓度加大或/和氨作用时间延长,细胞内成份开始出现增生修复现象,次级溶酶体数量增多、线粒体及内质网呈灶状增生。结论:观察结果提示,高氨可引起星形胶质细胞结构损伤基础上,星形胶质细胞出现增生修复现象。胶质细胞在高氟环境下所出现的变化可能是氨中毒的主要后果之一。
OBJECTIVE: To observe the effect of ammonia on the ultrastructure of astrocytes under in vitro conditions. MATERIALS AND METHODS: The high NH 4 + environment was induced by NH_4CI. The ultrastructural changes of the astrocytes cultured in vitro were observed. Results: In high NH 4 + environment, astrocytes showed the first damage: the decrease of electron density of cells, the swelling of organelles such as mitochondria and endoplasmic reticulum, the formation of many vacuoles in the cytoplasm with reduced electron density area. Later, with the increase of ammonia concentration or / and the prolonged action of ammonia, intracellular components began to proliferate and repair phenomenon, the number of secondary lysosomes increased, mitochondria and endoplasmic reticulum was focal hyperplasia. Conclusion: The results suggest that high ammonia can cause astrocyte structural damage, based on the phenomenon of astrocyte hyperplasia repair. Changes in glial cells in high-fluorine environment may be one of the main consequences of ammonia poisoning.
