目的建立高效液相法测定消脂胶囊中丹参酮ⅡA和丹酚酸B含量的方法。方法采用Grace Smart C18(4.6 mm×250 mm,5μm)色谱柱;丹参酮ⅡA以甲醇-水(75∶25)为流动相等度洗脱,波长为270 nm;丹酚酸B以甲醇-甲酸-水(40∶1∶59)为流动相等度洗脱,波长为286 nm;流速为1.0 m L·min-1;柱温为30℃;进样量为10μL。结果丹参酮ⅡA浓度在0.004~0.04 mg·L-1范围内呈良好的线性关系,平均回收率98.21%(RSD为2.817%,n=6);丹酚酸B在0.03~0.3 mg·L-1范围内呈良好的线性关系,平均回收率98.86%(RSD为2.119%,n=6)。结论该法快速,简便易行,结果可靠,专属性强,可作为消脂胶囊的质量控制指标。
Objective To establish a method for the determination of tanshinone ⅡA and salvianolic acid B in Xiaozhi capsule by HPLC. The method was performed on a Grace Smart C18 column (4.6 mm × 250 mm, 5 μm). Tanshinone ⅡA was eluted with mobile phase of methanol-water (75:25) at a wavelength of 270 nm. Salvianolic acid B (40: 1: 59) was used as eluent of mobile phase with the wavelength of 286 nm. The flow rate was 1.0 m L · min-1. The column temperature was 30 ℃. The injection volume was 10 μL. Results The concentration of tanshinone Ⅱ A showed a good linear relationship in the range of 0.004-0.04 mg · L-1 with an average recovery of 98.21% (RSD 2.817%, n = 6) and salvianolic acid B 0.03-0.3 mg · L -1 Within a good linear relationship, the average recovery was 98.86% (RSD 2.119%, n = 6). Conclusion The method is rapid, simple and easy to operate with reliable results and high specificity. It can be used as quality control index of Xiaozhi capsule.
