目的检测人类细胞表面糖蛋白2(SCUBE2)在乳腺癌组织中的表达情况,探讨SCUBE2表达与乳腺癌患者临床病理特征及乳腺癌关键基因的相关性。方法应用液相芯片基因检测技术检测152例乳腺癌组织样本SCUBE2、Bcl-2、Survivin及PTEN基因m RNA表达,分析上述指标与乳腺癌患者临床病理学资料及它们之间的相关性。结果在乳腺癌中SCUBE2 m RNA的表达与ER、PR及Ki67状态密切相关(P<0.05);进一步分析结果显示,中—高表达SCUBE2的患者Ki67活跃程度明显低于低表达SCUBE2的患者(P<0.05)。Survivin m RNA的表达与Ki67状态密切相关(P<0.05);PTEN m RNA表达与ER及PR状态密切相关(P<0.05);Bcl-2 m RNA表达与淋巴结转移、ER、PR及Ki67状态密切相关(P<0.05)。Spearsman等级相关法分析表明,SCUBE2表达与Bcl-2及PTEN表达呈正相关(r=0.434、0.348,P<0.01),而SCUBE2表达与Survivin表达呈负相关(r=-0.249,P<0.01)。结论在乳腺癌中SCUBE2表达与肿瘤增殖密切相关,可能是乳腺癌重要预后指标。
Objective To detect the expression of human cell surface glycoprotein 2 (SCUBE2) in breast cancer tissues and to investigate the correlation between the expression of SCUBE2 and the clinicopathological characteristics and the key genes of breast cancer in breast cancer patients. Methods The mRNA expression of SCUBE2, Bcl-2, Survivin and PTEN in 152 breast cancer samples was detected by liquid-phase microarray. The above indexes were analyzed with the clinicopathological data of breast cancer patients and their correlation. Results The expression of SCUBE2 mRNA in breast cancer was closely related to the status of ER, PR and Ki67 (P <0.05). Further analysis showed that the Ki67 activity of patients with moderate-high expression of SCUBE2 was significantly lower than that of patients with low expression of SCUBE2 (P <0.05). The expression of Survivin m RNA was closely related to the status of Ki67 (P <0.05). The expression of PTEN m RNA was closely related to the status of ER and PR (P <0.05). The expression of Bcl-2 mRNA was closely related to lymph node metastasis and ER, PR and Ki67 Related (P <0.05). Spearsman rank correlation analysis showed that the expression of SCUBE2 was positively correlated with the expression of Bcl-2 and PTEN (r = 0.434,0.348, P <0.01), while the expression of SCUBE2 was negatively correlated with the expression of Survivin (r = -0.249, P <0.01). Conclusion The expression of SCUBE2 in breast cancer is closely related to tumor proliferation, which may be an important prognostic indicator of breast cancer.
