心肌细胞中存在阴离子交换蛋白(AE),其主要作用是转运Cl-/HCO3-,调节细胞内PH和细胞内[Cl-]。文献表明AE1、AE2、AE3在心肌细胞mRNA水平均有表达,但在蛋白水平上表达的主要有nAE1和cAE3。缺血条件下,心肌细胞膜上Cl-/HCO3-交换蛋白可激活,导致[Cl-]i增加与酸中毒,造成心肌细胞损伤。除去细胞外Cl-或运用Cl-阻断剂,多种原因诱导的再灌注性心律失常发生率显著降低,表现出较好的抗心律失常作用。
Anion-exchange protein (AE) is present in cardiomyocytes, and its main role is to transport Cl- / HCO3-, regulating intracellular pH and intracellular [Cl-]. Literature shows that AE1, AE2, AE3 mRNA expression in cardiac muscle cells are expressed, but the protein expression mainly nAE1 and cAE3. Under ischemic conditions, Cl- / HCO3- exchangeable protein on the myocardial cell membrane can be activated, leading to [Cl-] i increase and acidosis, resulting in myocardial cell injury. Removal of extracellular Cl-or use of Cl-blockers, a variety of causes induced reperfusion arrhythmia was significantly reduced, showing better anti-arrhythmic effect.
