应用聚合酶链反应(PCR)检测急性和慢性乙肝患者外周血单个核细胞(PBMC)中的HBV-DNA,阳性率为63.8%。其中,急肝患者阳性率为78.6%;慢迁肝患者为56.3%;慢活肝患者为57.1%。PBMC中HBV-DNA与血清HBV-DNA和e抗原有一致性,但HBeAg阴性者中也有部分患者的PBMC中有HBV-DNA存在。本法消除了血清中HBV-DNA和PBMC自身DNA的干扰,并且与传统分子杂交方法相比,工作量大大减少。
The positive rate of HBV-DNA in peripheral blood mononuclear cells (PBMC) of patients with acute and chronic hepatitis B was 63.8% by polymerase chain reaction (PCR). Among them, the positive rate of acute liver disease was 78.6%; slow-moving liver was 56.3%; slow-living liver was 57.1%. PBMC HBV-DNA and serum HBV-DNA and e antigen consistency, but there are some patients with HBeAg-negative PBMC also exist HBV-DNA. This method eliminates the interference of serum HBV-DNA and PBMC’s own DNA, and greatly reduces the workload compared with the traditional molecular hybridization method.
