目的探讨Rad51及端粒酶逆转录酶在胶质瘤中的表达及与预后的关系。方法应用免疫组织化学SP法检测106例胶质瘤中Rad51及hTERT的表达。结果 (1)Rad51在胶质瘤中表达的总阳性率为52.8%(56/106),Ⅱ、Ⅲ、Ⅳ级的阳性率分别为31.3%(10/32)、55.9%(33/59)、86.7%(13/15);(2)hTERT在胶质瘤中表达的总阳性率为54.7%(58/106),Ⅱ、Ⅲ、Ⅳ级的阳性率分别为37.5(12/32)、57.6%(34/59)、80.0%(12/15);(3)Rad51蛋白表达和hTERT表达呈正相关(r=0.5428,P﹤0.01);(4)Ⅱ、Ⅲ、Ⅳ级胶质瘤患者术后平均生存时间分别为32.8、20.1、10.7个月;Rad51和hTERT阴性表达与阳性表达者间患者平均生存时间的差异均有统计学意义(P﹤0.05)。结论 Rad51和hTERT的表达与胶质瘤的病理分级呈正相关,与患者的术后生存时间呈负相关,两者联合检测有助于胶质瘤的早期诊断及预后评估。
Objective To investigate the expression of Rad51 and telomerase reverse transcriptase in gliomas and its relationship with prognosis. Methods Immunohistochemical SP method was used to detect the expression of Rad51 and hTERT in 106 gliomas. Results (1) The positive rate of Rad51 expression in glioma was 52.8% (56/106), the positive rates of grade II, III and IV were 31.3% (10/32) and 55.9% (33/59) , 86.7% (13/15) respectively. (2) The positive rate of hTERT expression in gliomas was 54.7% (58/106), the positive rates of grade Ⅱ, Ⅲ and Ⅳ were 37.5 (12/32) 57.6% (34/59), 80.0% (12/15) respectively; (3) There was a positive correlation between Rad51 expression and hTERT expression (r = 0.5428, P <0.01) The mean survival time after surgery was 32.8, 20.1 and 10.7 months, respectively. The mean survival time of patients with negative expression of Rad51 and hTERT was significantly higher than that of positive expression (P <0.05). Conclusion The expression of Rad51 and hTERT is positively correlated with the pathological grade of glioma, and negatively correlated with the postoperative survival time. Combined detection of Rad51 and hTERT contributes to the early diagnosis and prognosis evaluation of glioma.
