目的 探讨米非司酮与三苯氧胺合用抗早孕的作用机理。 方法 放射免疫方法测定绒毛、蜕膜组织培养液中人绒毛膜促性腺激素 (hCG)、孕酮 (P)、雌二醇 (E2 )、6 酮 前列腺素F1α(6 Keto PGF1α)、血栓素B2 (TXB2 )、泌乳素 (PRL)以及肿瘤坏死因子α(TNF α)、白细胞介素 (IL) 2。 结果 米非司酮与不同浓度三苯氧胺 (10、10 0、10 0 0、10 0 0 0nmol/L)合用 48h与对照组相比 ,绒毛组织培养液中hCG、P、E2 及 6 Keto PGF1α水平显著下降 (P <0 .0 5,P <0 .0 1) ,TXB2 水平显著升高 (P <0 .0 5) ,6 Keto PGF1α/TXB2 比值显著下降 (P <0 .0 1) ;蜕膜组织培养液中E2 、6 Keto PGF1α、PRL水平显著下降 (P <0 .0 5,P <0 .0 1) ,TNF α水平显著升高 (P <0 .0 1)。米非司酮与三苯氧胺合用对蜕膜E2 、TNF α水平的影响比米非司酮单独应用显著增强 (P <0 .0 5,P <0 .0 1)。 结论 米非司酮与三苯氧胺合用可显著影响绒毛、蜕膜组织的分泌功能 ,增加蜕膜组织TNF α含量 ;对蜕膜E2及TNF α水平的影响大于米非司酮单独应用
Objective To investigate the mechanism of anti-pregnancy effect of mifepristone combined with tamoxifen. Methods Radioimmunoassay was used to determine the expression of human chorionic gonadotropin (hCG), progesterone (P), estradiol (E2), 6 Keto PGF1α, thromboxane B2 (TXB2), prolactin (PRL), and tumor necrosis factor alpha (TNF alpha), interleukin (IL) 2. Results Compared with the control group, the levels of hCG, P, E2 and 6 Keto PGF1α in chorionic villi tissue of mifepristone treated with different concentrations of tamoxifen (10, 10, 10 000, 10 000 nmol / L) (P <0.05), the level of TXB2 was significantly increased (P <0.05), the ratio of 6 Keto PGF1α / TXB2 was significantly decreased (P <0.01) The levels of E2, 6 Keto PGF1α and PRL in the tissue culture medium were significantly decreased (P <0.05, P <0.01), and TNFα levels were significantly increased (P <0.01). The combination of mifepristone and tamoxifen had a significant effect on decidual E2 and TNFα levels compared with mifepristone alone (P <0.05, P <0.01). Conclusion Combination of mifepristone and tamoxifen can significantly affect the secretion of villi and decidua and increase the level of TNFα in decidual tissue. The effect on decidual E2 and TNFα levels is greater than that of mifepristone alone
