目的:探讨不同年龄供体正常胃粘膜端粒长度的变化规律。方法:采用Southern杂交及图像分析技术,检测26例76岁以下不同年龄供体正常胃粘膜端粒限制性片段长度(Telomericrestrictionfrag-ment,TRF)。结果:正常胃粘膜TRF长度呈年龄递减趋势,60岁以上组TRF长度明显短于0~39岁、40~49岁及50~59岁组(4.1±1.2vs6.4±1.8、6.0±1.0、5.5±1.1,P<0.05),正常胃粘膜TRF长度与供体年龄呈明显负相关(r=-0.56,P=0.0028),端粒重复序列平均每年丢失约(41±12)bp碱基对。结论:TRF长度可以作为衡量胃粘膜细胞衰老的一种生物学标志
Objective: To investigate the changes of telomere length of normal gastric mucosa in different age groups. Methods: Southern blotting and image analysis were used to detect telomeric restriction fragment length (Telomeric Restriction Frag-ment, TRF) of 26 normal gastric mucosa of different ages under 76 years of age. Results: The TRF length of normal gastric mucosa showed a trend of decreasing age. The length of TRF in patients over 60 years old was significantly shorter than that of 0-39 years old, 40-49 years old and 50-59 years old group (4.1 ± 1.2 vs 6.4 ± 1.8 , 6.0 ± 1.0,5.5 ± 1.1, P <0.05). There was a negative correlation between TRF length of donor and gastric mucosa (r = -0.56, P = 0.0028 ), Telomere repeats lost on average about (41 ± 12) bp base pairs each year. Conclusion: The length of TRF can be used as a biomarker to measure the cellular senescence of gastric mucosa
