目的:研究8-溴-7-甲氧基白杨素(8-bromo-5-hydroxy-7-methoxychrysin,BrMC)诱导人急性T淋巴细胞白血病Jurkat细胞凋亡作用。方法:体外培养Jurkat细胞。FITC标记annexinⅤ/PI染色流式细胞术(FCM)分析细胞凋亡率。ELISA法测定细胞组蛋白/DNA碎片。二氯荧光素二乙酯(2’7’-dichlorofluoresein diacetate,DCFH-DA)荧光探针FCM检测细胞活性氧。结果:BrMC以剂量依赖方式诱导annexinⅤ阳性细胞和组蛋白/DNA碎片增加(P<0.05)。BrMC促进细胞活性氧生成,10 mmol/L N-乙酰半胱氨酸(NAC)预孵育能有效阻断Jurkat细胞活性氧生成,并减弱其诱导细胞凋亡效应。结论:BrMC具有诱导Jurkat细胞凋亡作用,其作用与促进细胞活性氧生成有关。
AIM: To investigate the apoptosis of human acute lymphoblastic leukemia Jurkat cells induced by 8-bromo-5-hydroxy-7-methoxychrysin (BrMC). Methods: Jurkat cells were cultured in vitro. FITC labeled annexin Ⅴ / PI staining flow cytometry (FCM) analysis of apoptosis rate. ELISA for determination of cellular histone / DNA fragments. Fluorescence probe FCM of 2’7’-dichlorofluoresein diacetate (DCFH-DA) was used to detect the cellular reactive oxygen species. Results: BrMC induced an increase in annexin Ⅴ positive cells and histone / DNA fragments in a dose-dependent manner (P <0.05). BrMC promoted the production of reactive oxygen species. Preincubation with 10 mmol / L NAC could effectively block the generation of reactive oxygen species (ROS) in Jurkat cells and attenuate the induction of apoptosis. Conclusion: BrMC can induce the apoptosis of Jurkat cells, and its effect is related to the promotion of reactive oxygen species production.
