目的建立以HPLC为基础的测定黄瓜中β-胡萝卜素的含量方法。方法采用Agilent Eclipse XDB-C18(5μm,4.6mm*250mm)色谱柱。流动相A为甲醇与乙腈的混合溶液,甲醇的含量为30%,流动相B为二氯甲烷,采用梯度洗脱,流速为1mL/min,柱温为35℃,检测波长为450nm,采用外标法定量。结果β-胡萝卜素在10μg-100μg?mL-1内线性关系良好,线性方程为y=8272.6x+788.71,R=0.9995(n=5),平均回收率为92.31%,RSD=1.2%(n=6)。结论:该测定黄瓜中β-胡萝卜素的反相高效液相色谱方法简便且准确可靠。
Objective To establish a HPLC-based method for the determination of β-carotene in cucumber. Methods Agilent Eclipse XDB-C18 (5 μm, 4.6 mm × 250 mm) columns were used. The mobile phase A was a mixed solution of methanol and acetonitrile. The content of methanol was 30%. The mobile phase B was dichloromethane. The gradient elution was used with a flow rate of 1 mL / min, a column temperature of 35 ℃ and a detection wavelength of 450 nm. Standard method of quantitative.
