目的:探讨氧载体对茯苓菌液态深层发酵的调控效应。方法:采用单因素试验设计分别考察氧载体种类、浓度和添加时间对茯苓菌液态深层发酵的影响,进而确定氧载体添加策略,并分析氧载体对茯苓菌在10 L生物反应器中的代谢调控过程。结果:氧载体最佳添加方案为接种48 h后添加1.0%(V/V)吐温-80,此时10 L反应器中茯苓菌细胞干重可达13.43 g/L,胞外多糖和茯苓酸产量分别为8.58 g/L、989.52μg/L,明显高于未添加氧载体发酵体系。结论:吐温-80可显著提高液态深层发酵体系中茯苓菌细胞生长、胞外多糖合成及茯苓酸积累水平,为解决高等真菌液态发酵过程中因溶氧限制引起的菌体生长和产物抑制问题提供了参考。
Objective: To investigate the regulatory effect of oxygen carrier on liquid deep fermentation of Poria cocos. Methods: The single factor experiment design was used to investigate the effect of oxygen carrier type, concentration and adding time on the liquid deep fermentation of Poria cocos, and then to determine the strategy of oxygen carrier addition, and to analyze the metabolic regulation of Poria cocos infection in 10 L bioreactor process.
