Serum glycosyltransferases and carbohydrases are associated with the presence of malignant tumors and their degree of metastasis. A recent report that a galactosyltransferase isozyme appears to be tumor-specific. This article determined and compared the levels of these enzymes in entero-intestinal, colon cancer-cultured cells, surgically-operated colon cancer, and adjacent normal tissues. Three galactosyltransferases, one N-acetylgalactosamine transferase and two sialyltransferases were tested for activity, and siallase activity was measured. Surgical specimens were placed in 5 vol of cold PBS, homogenized in a homogenizer for 1 minute, sonicated for 15 seconds and centrifuged, all of which were performed rapidly at 4 ° C. The cells were washed five times with PBS, centrifuged, and the cells were suspended in methhenazepine buffer and sonicated. The extract was used directly as a glycosyltransferase or glycosidase activity assay or in a solution containing 0.1% Triton X- 100 solution, after sonic treatment, for isoelectric focusing electrophoresis points
