目的 :探讨加热抗原修复对甲醛固定石蜡包埋胃癌组织中内源性生物素样分子活性的影响。方法 :采用煮沸法、免疫组织化学 SP染色法及抗生物素蛋白 -生物素依次处理法 ,对 10例甲醛固定石蜡包埋胃癌标本内的内源性生物素样分子的活性进行检测。结果 :1经甲醛固定和石蜡包埋的胃癌组织中内源性生物素样分子被灭活 ;2加热抗原修复显著增强内源性生物素样分子的活性 ,同时 ,内源性生物素样分子仅见于癌细胞胞浆中而未见于间质细胞 ;3抗生物素蛋白 -生物素依次处理法可封闭内源性生物素样分子。结论 :加热抗原修复可显著增强甲醛固定石蜡包埋胃癌组织中内源性生物素样分子的活性 ,而为避免其对正常免疫组织化学染色的影响 ,可采用抗生物素蛋白 -生物素依次处理法或运用非生物素检测系统。
Objective: To investigate the effect of heated antigen retrieval on the activity of endogenous biotin-like molecules in formalin-fixed paraffin-embedded gastric cancer tissues. Methods: The activity of endogenous biotin-like molecules in 10 formaldehyde-fixed paraffin-embedded gastric cancer specimens was detected by boiling method, immunohistochemical SP staining and avidin-biotin sequential treatment. Results: 1 Endogenous biotin-like molecules were inactivated by formalin-fixed and paraffin-embedded gastric cancer tissues. 2 Heated antigen repair significantly enhanced the activity of endogenous biotin-like molecules. At the same time, endogenous biotin-like molecules Only found in the cytoplasm of cancer cells but not found in stromal cells; 3 avidin-biotin sequential processing method can be closed endogenous biotin-like molecules. CONCLUSION: Heated antigen repair can significantly enhance the activity of endogenous biotin-like molecules in formalin-fixed paraffin-embedded gastric cancer tissues, and in order to avoid its impact on normal immunohistochemical staining, avidin-biotin sequential treatment Law or the use of non-biotin detection system.
