目的:运用转基因Kkay2型糖尿病小鼠为模型,观察用辛开苦降法传统煎剂(辛开苦降第1组)、辛开苦降法颗粒剂(辛开苦降第2组)干预对Kkay 2型糖尿病小鼠胰岛素敏感性及血脂的疗效。方法:10周龄雄性初发2型糖尿病KKay小鼠36只按血糖值随机分为4组:模型组、罗格列酮组(0.67 g·kg-1·d-1)、辛开苦降煎剂组(8.7 g·kg-1·d-1)、辛开苦降颗粒剂(8.7 g·kg-1·d-1)。每天灌胃给药1次,4周后测定血糖、空腹血糖、胰岛素、血脂,计算胰岛素敏感指数。结果:给药4周后,与正常组相比,模型组胰岛素敏感指数明显降低(P<0.01),空腹血糖、甘油三酯、总胆固醇等明显升高。辛开苦降煎剂、辛开苦降颗粒剂与模型组相比,胰岛素敏感指数高于模型组(P<0.01),空腹血糖值低于模型组(P<0.01);甘油三酯、总胆固醇均低于模型组,辛开苦降传统煎剂和辛开苦降法颗粒剂之间并无差异。结论:辛开苦降法可改善初发2型糖尿病KKay小鼠的胰岛素抵抗,增加胰岛素的敏感性从而改善血糖水平,并能降低血脂水平,且煎剂和颗粒剂疗效相当。
OBJECTIVE: To investigate the effects of Xinkai biting method (Xinkai bitter down group 1), Xinkai biting method granule (Xinkai bitter down group 2) Insulin sensitivity and blood lipids in Kkay type 2 diabetic mice. Methods: Thirty-six male KKay mice with 10-week-old type 2 diabetes mellitus were randomly divided into 4 groups: model group, rosiglitazone group (0.67 g · kg-1 · d-1) Decoction group (8.7 g · kg-1 · d-1) and Xinkai biting granules (8.7 g · kg-1 · d-1). Gavage once a day, 4 weeks after the determination of blood glucose, fasting blood glucose, insulin, lipids, insulin sensitivity index calculation. Results: Compared with the normal group, the insulin sensitivity index of the model group decreased significantly (P <0.01) and the fasting blood glucose, triglyceride and total cholesterol were significantly increased after 4 weeks. Xinkai bitter decoction, Xin Kai Kuishang granules compared with the model group, the insulin sensitivity index was higher than the model group (P <0.01), fasting blood glucose was lower than the model group (P <0.01); triglyceride, total Cholesterol were lower than the model group, Xin Kai bitter traditional decoction and Xin Kai Kui Shaofan granules no difference between. Conclusion: Xinkai biting down method can improve insulin resistance of newly diagnosed type 2 diabetes KKay mice, increase insulin sensitivity to improve blood sugar level, and can reduce blood lipid levels, and the decoction and granules efficacy.
