目的探讨经颈动脉途径肾动脉插管造影骨髓间充质干细胞(MSCs)移植对慢性阿霉素肾病大鼠肾脏的影响,为临床干细胞移植治疗慢性肾脏病及其给药途径选择提供参考。方法 50只雄性SD大鼠作为实验动物。36只采用经尾静脉注射阿霉素制作慢性肾病大鼠模型,造模后随机分为3组(每组12只):(1)阿霉素肾衰模型对照组(ADR组);(2)MSCs移植组(A组),取MSCs悬浮液经肾动脉插管造影输入大鼠右肾;(3)生理盐水组(B组),以A组同样方法将生理盐水输入大鼠右肾。2只用于骨髓MSCs的制备。其余12只作为空白对照组(N组)。干细胞移植后第1天和第7天分别测定各组大鼠肾功能(尿素氮、肌酐)和血红蛋白含量,第7天光镜下观察各组大鼠肾脏病理改变。结果 (1)ADR组、A组、B组与N组比较血尿素氮、肌酐明显升高(P<0.05),血红蛋白含量显著降低(P<0.05)。(2)第1天,与ADR组比较,A组、B组血尿素氮、肌酐和血红蛋白含量无统计学差异(P均>0.05);第7天时,与ADR组比较,B组血尿素氮、肌酐和血红蛋白含量无统计学差异(P均>0.05),A组血尿素氮、肌酐明显降低(P均<0.05),血红蛋白含量明显升高(P<0.05)。(3)光镜下大鼠肾脏病理改变,B组肾小球、肾小管、肾间质损伤程度与ADR组相当,A组肾间质炎症改变程度轻且范围小。结论移植MSCs有利于慢性阿霉素肾病大鼠肾脏损伤的修复,经颈动脉肾动脉插管造影是可行的途径。
Objective To investigate the effects of transarterial renal artery cannulation (MSA) on the kidneys of chronic adriamycin-induced nephropathy rats and provide a reference for clinical stem cell transplantation in the treatment of chronic kidney disease and its route of administration. Methods Fifty male SD rats were used as experimental animals. Thirty-six rats were randomly divided into 3 groups (12 in each group): (1) Adriamycin-induced renal failure model group (ADR group); (2) ) Group (group A). The MSCs suspension was injected into the right kidney of rats through renal artery cannulation angiography. (3) The saline group (group B), the same method as group A, saline was injected into the right kidney of rats. 2 only for the preparation of bone marrow MSCs. The remaining 12 as a blank control group (N group). The renal function (urea nitrogen, creatinine) and hemoglobin content of rats in each group were measured on the 1st day and the 7th day after stem cell transplantation. The pathological changes of the kidneys of the rats in each group were observed under light microscope on the 7th day. Results (1) Blood urea nitrogen and creatinine were significantly increased (P <0.05), hemoglobin content was significantly lower in ADR group, A group and B group than in N group (P <0.05). (2) Compared with ADR group, there was no significant difference in blood urea nitrogen, creatinine and hemoglobin between group A and group B on the first day (all P> 0.05). On the seventh day, compared with the ADR group, the blood urea nitrogen , Creatinine and hemoglobin content were not significantly different (P> 0.05). Blood urea nitrogen and creatinine were significantly decreased in group A (all P <0.05) and hemoglobin content was significantly increased (P <0.05). (3) Under the light microscope, pathological changes of rat kidney were observed. The degree of glomerular, tubular and renal interstitium injury in group B was similar to that in ADR group. The severity of renal interstitial inflammation in group A was mild and the range was small. Conclusion Transplantation of MSCs is beneficial to the repair of renal injury in chronic adriamycin-induced nephropathy rats. Carotid artery renal artery catheterization is a feasible approach.
