目的探讨超极化激活环核苷酸门控阳离子通道2蛋白(HCN2)和细胞膜锚蛋白(ANK2)在心源性猝死(SCD)病人窦房结的表达水平。方法收集因心源性疾病猝死的个体心脏14例作为SCD组;选择无心脏疾病的死亡个体心脏标本13例作为非SCD组。准确切取心脏窦房结组织,石蜡包埋,部分HE染色,采用免疫组织化学的方法分别测定两组HCN2和ANK2蛋白表达水平。结果 SCD组HCN2在窦房结组织中呈棕黄色颗粒,非均质分布在核旁;SCD组ANK2稀疏分布于窦房结细胞浆内。同非SCD组相比,SCD组HCN2蛋白表达水平上调和ANK2蛋白表达水平下调(0.29±0.03vs 0.16±0.07;0.09±0.06vs 0.23±0.06,P均<0.05)。结论 SCD患者窦房结HCN2基因表达水平上调,ANK2基因表达水平下调。
Objective To investigate the expression of HCN2 and ANK2 in sinus node in patients with sudden cardiac death (SCD). Methods 14 individuals with sudden cardiac death were collected as SCD group. Thirteen death cardiac samples from non-cardiac patients were selected as non-SCD group. The heart sinus node tissue, paraffin-embedded, and partially HE staining were obtained. The expression of HCN2 and ANK2 protein in the two groups were determined by immunohistochemistry. Results HCN2 in SCD group showed a yellowish-yellow granules in the sinus node tissue, heterogeneously distributed in the nucleus; ANK2 in SCD group was sparsely distributed in the cytoplasm of sinoatrial node. Compared with non-SCD group, HCN2 protein expression and ANK2 protein expression in SCD group were down-regulated (0.29 ± 0.03 vs 0.16 ± 0.07; 0.09 ± 0.06 vs. 0.23 ± 0.06, P <0.05 respectively). Conclusion The expression of HCN2 gene in the sinus node and the expression of ANK2 in SCD patients are up-regulated.
