目的:探讨原花青素(PC)对反式脂肪酸(TFA)染毒小鼠睾丸组织及其激素损伤的修复作用。方法:50只昆明种雄性小鼠随机等分为5组:溶剂对照组、TFA染毒组、TFA染毒+低、中、高剂量PC保护组。TFA染毒组小鼠按100.0 mg/(kg·bw)剂量灌胃TFA;低、中、高剂量PC保护组小鼠分别按25、50、100 mg/(kg·bw)剂量灌胃PC溶液。对照组灌胃生理盐水,其它各组均上午灌胃TFA、保护组下午灌胃PC溶液,每天1次,实验时间均为90 d。实验毕处死动物取血及双侧睾丸测定相关指标。结果:TFA染毒组睾丸组织形态结构损伤严重,出现基底膜模糊不清、核间隙增宽,支持细胞水肿、破裂,精子数少等病变;给予不同剂量PC保护后睾丸的结构受损较TFA染毒组有明显所改善,以高剂量PC改善明显;TFA染毒组小鼠血清FSH、LH、T值及睾丸中SOD活性均低于对照组,而MDA含量高于对照组(P<0.05);经不同剂量PC保护后小鼠血清FSH、LH、T值及睾丸中SOD活性均高于对照组,而MDA含量低于对照组(P<0.05)。结论:过量摄入TFA可致小鼠睾丸及雄激素受损;经一定剂量的PC保护后其受损的睾丸结构及雄激素有修复作用,其作用机制可能与抗氧化损伤有关。
Objective: To investigate the effect of proanthocyanidins (PC) on the repair of testis and its hormones in mice exposed to trans fatty acids (TFA). Methods: Fifty Kunming male mice were equally divided into five groups: solvent control group, TFA-treated group, TFA-treated + low, middle and high dose PC-protection group. The mice in TFA group were administered with TFA at the dose of 100.0 mg / (kg · bw), while the mice in low, middle and high doses of PC group were administered with 25, 50, 100 mg / (kg · bw) . The control group was given normal saline, and the other groups were fed with TFA in the morning. The protection group was given intragastric administration of PC solution once a day for 90 days. Experimental completion of animal blood and bilateral testis related indicators. Results: The testicular tissue morphology of TFA group was severely damaged. There was unclear basement membrane, broadening of nuclear gap, supporting cell edema, rupture and few sperm count. The structure of testis damaged by TFA exposure was better than that of TFA The levels of FSH, LH and T in serum and the activity of SOD in testis were significantly lower than those in control group, while the content of MDA in TFA group was higher than that in control group (P <0.05) ); The serum FSH, LH, T and the activity of SOD in the testis were higher than those in the control group, while the content of MDA in the testis was lower than that in the control group (P <0.05). CONCLUSION: TFA overexpresses the testis and androgen in mice. After a certain dose of PC protection, the damaged testicular structure and androgen may have a repair effect, which may be related to the anti-oxidation injury.
