以芍药不同花发育时期和不同组织器官为研究材料,利用qPCR技术检测8个内参基因的表达水平,并利用geNorm,NormFinder,Bestkeeper和RefFinder软件综合评价它们的表达稳定性。结果表明,分析芍药花瓣不同发育时期和不同组织的基因表达时可选用UBQ,GAPDH和ACTIN作为内参基因,而PP2A和SAMS不适于作为内参基因。PlF3H基因的相对表达水平证实,在芍药花瓣不同发育时期,利用UBQ和GAPDH两个表达最稳定的组合即可获得精确的基因表达结果。
The expression levels of eight reference genes were detected by qPCR at different developmental stages and different tissues and organs of Paeonia lactiflora. GeNorm, NormFinder, Bestkeeper and RefFinder software were used to evaluate their expression stability. The results showed that UBQ, GAPDH and ACTIN could be used as internal reference genes when analyzing the gene expression of peony flower petals at different developmental stages and different tissues, while PP2A and SAMS were not suitable as internal reference genes. The relative expression level of PlF3H gene confirmed that at the different developmental stages of peony petals, accurate gene expression results can be obtained by using the two most stable combinations of UBQ and GAPDH.
