目的:研究膜联蛋白A2(AnnexinA2,ANXA2)、膜联蛋白A4(AnnexinA4,ANXA4)在人体胃腺癌(gastric adenocarcinoma cells,GAC)的表达,探讨其表达与GAC生物学行为的关系及两种蛋白的相关性.方法:免疫组织化学SP法检测组织芯片中75对配对的GAC和其相应癌旁组织ANXA2和ANXA4的表达.结果:在GAC和癌旁组织中ANXA2、ANXA4的表达分别为:33.1%(25)vs1.3%(1)、68.0%(51)vs25.5%(13),二者在GAC的表达显著高于癌旁组织(x2=24.448,P=0.000;x2=39.353,P=0.000);ANXA2和ANXA4在GAC表达呈正相关(r=0.335,P=0.003);两种蛋白的表达与GAC的浸润深度、淋巴结转移、TNM分期和分化程度有关(P<0.05),与年龄、性别和远处转移无关(P>0.05);肿瘤大小与ANXA2表达有关(P<0.01),与ANXA4表达无关(P>0.05).结论:ANXA2和ANXA4的表达上调可能共同参与了GAC的发生、发展、浸润和转移,可作为临床预测GAC预后的标志物和治疗靶点.
OBJECTIVE: To study the expression of Annexin A2 and ANXA4 in human gastric adenocarcinoma cells (GAC) and to investigate their relationship with the biological behavior of GAC and to investigate the relationship between the expression of two proteins .Methods: Immunohistochemical SP method was used to detect the expression of ANXA2 and ANXA4 in 75 matched paired GACs and their corresponding paracancerous tissues.Results: The expressions of ANXA2 and ANXA4 in GAC and paracancerous tissues were 33.1 (25) vs1.3% (1), 68.0% (51) vs25.5% (13), both of which were significantly higher in GAC than in paracancerous tissues (x2 = 24.448, P = 0.000; x2 = 39.353, P = 0.000). There was a positive correlation between the expression of ANXA2 and ANXA4 in GAC (r = 0.335, P = 0.003). The expression of both proteins was related to the depth of invasion, lymph node metastasis, TNM stage and differentiation (P <0.01), but had no correlation with ANXA4 expression (P> 0.05) .Conclusion: The up-regulation of ANXA2 and ANXA4 may participate in the GAC Occurrence, development, invasion and metastasis, which can be used as a marker and target of clinical prediction of GAC prognosis.
