总结国外合成致死作用在抗肿瘤药物筛选中的最新研究进展。采用PubMed数据库检索系统,以“合成致死作用、抗肿瘤、药物筛选”等为关键词,检索1990年至今合成致死作用在抗肿瘤药物发现中的作用的文献。合成致死作用研究主要集中在PARP抑制剂和BRCA1/2缺失、MSH和DNA聚合酶、VHL基因、PTEN基因缺失、RAS和MYC基因的失调等。合成致死作用在抗肿瘤药物筛选中具有广泛应用,有望作为新一代抗肿瘤药物的筛选方法。
The recent research progress of synthetic lethality in anti-cancer drug screening was summarized. Using the PubMed database search system, we searched the literature on the role of synthetic lethality in the discovery of anti-tumor drugs from the perspective of “synthetic lethality, anti-tumor, drug screening” and so on. Synthetic lethal effects mainly focused on the PARP inhibitor and BRCA1 / 2 deletion, MSH and DNA polymerase, VHL gene, PTEN gene deletion, RAS and MYC gene disorders. Synthetic lethal effect in the screening of anti-cancer drugs has a wide range of applications, is expected as a new generation of anti-tumor drugs screening method.
