目的:从宁肠汤对束缚应激致腹泻大鼠小肠运动功能及血浆酪神经肽(NPY)的影响,探讨宁心安神法对腹泻型肠易激综合征(IBS)治疗的作用机理。方法:将72只雄性大鼠随机分为正常对照组、模型对照组、得舒特组、宁肠汤低、中、高剂量组,采用williams改良方法建立束缚应激+适度夹尾刺激致大鼠泄泻模型,观察宁肠汤对大鼠小肠运动功能的影响;并以酶联免疫分析法检测各组大鼠血浆中NPY的变化。结果:模型对照组大鼠小肠墨汁推进率较正常对照组明显提高(P<0.05),而各治疗组较模型对照组明显下降(P<0.05);模型组大鼠血浆NPY水平较正常对照组明显升高(P<0.01),大剂量宁肠汤能明显降低已经升高的大鼠血浆NPY含量。结论:宁肠汤可调节束缚应激致腹泻大鼠的小肠运动功能,大剂量宁肠汤能明显降低已经升高的大鼠血浆NPY含量。
Objective: To investigate the effect of Ningxin Anshen Therapy on the treatment of diarrhea-predominant irritable bowel syndrome (IBS) by observing the effect of Ningchang Decoction on small intestine motility and the level of plasma neuropeptide (NPY) in sham-dependent diarrhea-induced rats. Methods: Seventy-two male rats were randomly divided into normal control group, model control group, dextrose group, and ningchang decoction low, medium and high dose group. Williams modified method was used to establish restraint stress Rat diarrhea model to observe the effect of Ningchang Decoction on small intestine motility of rats; and the change of plasma NPY in each group was detected by enzyme-linked immunosorbent assay. Results: Compared with the normal control group, the promoting rate of small intestine ink in the model control group was significantly higher than that in the normal control group (P <0.05), while the levels of plasma NPY in the model group were significantly lower than those in the normal control group (P <0.01). Large dose of Ningchang Decoction can significantly reduce the plasma NPY content in rats. Conclusion: Ningchang Decoction can regulate small intestine motility of rats with restraint stress and high dosage of Ningchang Decoction can significantly reduce the plasma NPY content of rats.
