目的:观察碧血胶囊对大鼠急性心肌梗塞(AMI)后血流动力学变化及心功能的影响,探讨其预防AMI的机制。方法:结扎大鼠冠状动脉左前降支造成AMI模型,成年雄性SD大鼠50只随机分为6组:假手术组、急性心肌梗塞模型(AMI)组、通心络胶囊组、碧血胶囊高、低剂量组、地尔硫卓组。观察药物对各组大鼠心率(HR)、左心室收缩峰压均值(LVSP)、左心室舒张压均值(LVDP)、左心室收缩压最大上升速率(+dp/dtmax)、左心室舒张压最大下降速率(-dp/dtmax)、左心室开始收缩至达到室内压最大上升速率时间(t-dp/dtmax)、心律失常、血清游离脂肪酸(FFA)和天冬氨酸氨基转移酶(AST)的影响。结果:大鼠AMI后,HR、mlVSP、±dp/dtmax明显降低,LVDP,AST明显升高(P<0.05),并且随着缺血时间的延长,各指标呈逐渐恢复的趋势。与AMI组相比,碧血胶囊高(968mg/kg,结扎后120分钟)、低剂量组(484mg/kg,结扎时)可显著提高LVSP,碧血胶囊高(结扎时及结扎后120分钟时)、低剂量组(结扎后30分钟)可显著提高+dp/dtmax,碧血胶囊高(结扎时及结扎后30、120分钟时)可显著提高-dp/dtmax(P<0.05),同时碧血胶囊高、低剂量组均能降低血清AST及心律失常的发生次数(P<0.05)。结论:碧血胶囊能显著改善左室收缩和舒张功能,减轻大鼠心肌缺血后心功能的下降和降低血清AST含量,对AMI后血流动力学改变有明显的改善作用,这可能是其保护心肌梗死的机制之一。
Objective: To observe the effect of Bi-Xue Capsule on hemodynamics and cardiac function after acute myocardial infarction (AMI) in rats, and to explore its mechanism of preventing AMI. Methods: AMI model was induced by ligation of the left anterior descending coronary artery in rats. Fifty adult male Sprague-Dawley rats were randomly divided into 6 groups: sham-operated group, AMI group, Tongxinluo capsule group, Low-dose group, diltiazem group. The effects of drugs on heart rate (HR), left ventricular systolic pressure (LVSP), left ventricular diastolic pressure (LVDP), maximal rate of left ventricular systolic pressure (+ dp / dtmax) and left ventricular diastolic pressure (-dp / dtmax), left ventricular contractions to t-dp / dtmax, arrhythmia, serum free fatty acid (FFA), and aspartate aminotransferase influences. Results: After AMI, HR, mlVSP and ± dp / dtmax were significantly decreased, LVDP and AST were significantly increased (P <0.05), and with the extension of ischemic time, the indicators showed a gradual recovery. Compared with the AMI group, the BIAO capsule (968mg / kg, 120 minutes after ligation) and the low dose group (484mg / kg, ligation) significantly increased the LVSP and BIOU capsules (at ligation and 120 minutes after ligation) In the low dose group (30 minutes after ligation), the level of + dp / dtmax increased significantly (P <0.05) at the time of ligation and at 30,120 minutes after ligation. At the same time, Low-dose group can reduce the number of serum AST and arrhythmia (P <0.05). CONCLUSION: Bi-Xue Capsule can significantly improve left ventricular systolic and diastolic function, reduce cardiac function decline and serum AST content after myocardial ischemia in rats, and has a significant effect on hemodynamic changes after AMI, which may be its protection One of the mechanisms of myocardial infarction.
