目的:对痛风泰颗粒治疗大鼠急性痛风性关节炎的镇痛及抗炎作用进行实验研究。方法:将SD大鼠120只随机分为6组,即空白对照组(A组)、模型对照组(B组)、双氯芬酸组(C组)、痛风泰低剂量组(D组)、痛风泰中剂量组(E组)、痛风泰高剂量组(F组),每组20只,采用Coderre TJ方法造模后,分别予灌服生理盐水,双氯芬酸溶液及不同浓度的痛风泰溶液,观察其对关节周径的影响,并分别于治疗第3、7天后取关节滑膜进行组织匀浆,用ELISA酶联免疫法,检测关节滑膜组织中细胞因子5-HT的含量变化。结果:①双氯芬酸组、痛风泰中剂量组及痛风泰高剂量组在用药72 h后关节肿胀明显减轻,较造模后比较,差异有显著意义(P<0.01)。②痛风大鼠关节滑膜中疼痛介质5-HT的含量变化:治疗3 d后,与模型对照组比较,各治疗组5-HT含量明显降低,差异有统计学意义(P<0.05);经治疗7 d后,与空白对照组比较,双氯芬酸、痛风泰中剂量及痛风泰高剂量组均未见明显差异,P>0.05。结论:痛风泰颗粒能减轻MSUC所致急性痛风性关节炎关节肿胀程度,能降低5-HT浓度,减少疼痛介质的释放,较好地发挥镇痛作用。
Objective: To study the analgesic and anti-inflammatory effects of Goutitai Granules in treating acute gouty arthritis in rats. Methods: One hundred and twenty SD rats were randomly divided into 6 groups: blank control group (group A), model control group (group B), diclofenac group (group C), goutytai low dose group (group D) (Group E) and high dose of Gout group (group F), with 20 rats in each group. After modeling by Coderre TJ method, the rats were given saline, diclofenac solution and goutystat solution On the joint circumference, and synovial synovial tissue was harvested on the 3rd and 7th day after treatment, respectively. The content of cytokines 5-HT in the synovial tissue was detected by enzyme-linked immunosorbent assay (ELISA). Results: ① The swelling of joint in diclofenac group, middle dose of Goutytai group and high dose of Goutytai group were significantly relieved after 72 hours of treatment, and there was significant difference between the two groups (P <0.01). (2) Changes of 5-HT content in pain synovium of gout rats: After 3 days of treatment, the content of 5-HT in each treatment group was significantly lower than that of model control group (P <0.05) After 7 days of treatment, compared with the blank control group, Diclofenac, gout and middle-dose and high-dose group were no significant difference, P> 0.05. CONCLUSION: Tongfutai Granule can reduce the degree of swelling of joint caused by acute gouty arthritis caused by MSUC, reduce the concentration of 5-HT, reduce the release of pain medium and exert the analgesic effect better.
