【目的】建立单面针药材中木兰花碱的含量测定方法,为该药材质量评价方法的建立提供基础。【方法】采用高效液相色谱法,色谱柱为Kromasil C18(4.6 mm×250 mm,5μm);流动相为乙腈-0.1 mol/L磷酸二氢钠(体积比为11∶89);检测波长268 nm;柱温25℃。【结果】木兰花碱在20~300μg/mL范围内线性关系良好,r为0.999 9;理论塔板数不低于25 000;样品中木兰花碱检测无干扰,与邻峰分离度大于1.5,样品含量在0.74~6.61 mg/g之间,平均含量为2.99 mg/g,加样回收率为97%以上,sR为0.81%。【结论】所建方法简单可行,重复性好,可用于单面针药材的质量控制。
【Objective】 To establish a method for determination of Magnoliaceae in one-sided needles and provide the basis for establishing the quality evaluation method of this herb. 【Method】 High performance liquid chromatography (HPLC) was used with Kromasil C18 column (4.6 mm × 250 mm, 5 μm) as the mobile phase. The mobile phase consisted of acetonitrile-0.1 mol / L sodium dihydrogen phosphate nm; column temperature 25 ℃. 【Result】 The results showed that there was a good linear relationship between magnolol and alkaloids in the range of 20 ~ 300μg / mL, r was 0.999 9; the number of theoretical plates was not less than 25 000; The sample contents ranged from 0.74 to 6.61 mg / g, with an average content of 2.99 mg / g. The recoveries of samples were above 97% with sR of 0.81%. 【Conclusion】 The proposed method is simple and feasible with good reproducibility and can be used for the quality control of single-sided needles.
