[目的]探讨胃宁方对胃癌前病变(PLGC)大鼠转化生长因子β(TGF-β)/Smad信号通路的影响。[方法]60只大鼠随机分为6组:空白对照(空白)组,模型组,维甲酸组,胃宁方低、中、高剂量(低、中、高剂量)组,每组10只,采用以N-甲-N-硝基-亚硝基胍(MNNG)为主的4因素联合造模法建立PLGC大鼠模型。维甲酸组给予维甲酸水溶剂1.0 g/kg;低、中、高剂量组分别给予胃宁方生药浓度为1.0、2.0、3.0 g/ml的煎剂,10 ml/kg,均每日1次灌胃。12周后观察各组大鼠一般情况、胃黏膜病理组织学变化,以及对TGF-β1、Smad4 mRNA表达和细胞外信号调节激酶(ERK1)水平的影响。[结果]胃宁方各剂量组大鼠活动、饮食等一般情况好转;模型组与空白组比较,TGF-β1、ERK1表达水平明显增高,Smad4表达水平明显降低;低、中、高剂量组与模型组比较,TGF-β1、ERK1表达水平下调,Smad4表达上调,差异均有统计学意义(P<0.05,<0.01)。[结论]胃宁方可改善PLGC大鼠胃黏膜病理组织状况,下调TGF-β1mRNA表达及ERK1水平,上调Smad4 mRNA表达。
[Objective] To investigate the effect of Weining Fang on TGF-β / Smad signal pathway in gastric precancerous lesions (PLGC) rats. [Method] 60 rats were randomly divided into 6 groups: control group (blank), model group, retinoic acid group and Weining Fang low, medium and high dose (low, medium and high dose) The model of PLGC was established by 4-factor combined modeling with N-methyl-N-nitro-nitrosoguanidine (MNNG) as the mainstay. Retinoic acid group given retinoic acid water solvent 1.0 g / kg; low, medium and high dose groups were given Weining Fang crude drug concentration of 1.0,2.0,3.0 g / ml of decoction, 10 ml / kg, 1 times a day Gavage. After 12 weeks, the general situation of rats in each group, the histopathological changes of gastric mucosa and the effects of TGF-β1, Smad4 mRNA expression and extracellular signal-regulated kinase (ERK1) levels were observed. [Results] The general conditions such as activity and diet of each dose group of Weiningnfang improved. Compared with the blank group, the expression of TGF-β1 and ERK1 increased significantly and the expression of Smad4 decreased obviously. Compared with the blank group, Compared with model group, the expression of TGF-β1, ERK1 was down-regulated and the expression of Smad4 was up-regulated. The differences were statistically significant (P <0.05, <0.01). [Conclusion] Weining Decoction can improve gastric mucosal pathological conditions, down-regulate the expression of TGF-β1 mRNA and ERK1, and up-regulate the expression of Smad4 mRNA in PLGC rats.
