目的:观察黄芩茎叶总黄酮(SSTF)对单纯高甘油三酯血清(HTG)刺激的血管平滑肌细胞(VSMCs)增殖的抑制作用及可能的分子机制。方法:体外培养大鼠胸主动脉VSMCs,HTG诱导其增殖,观察SSTF对细胞增殖、细胞周期、CO含量、血红素氧化酶-1(HO-1)和ERK/p-ERK蛋白表达的影响。结果:500 mg.L-1的SSTF可明显抑制HTG诱导的VSMCs增殖,增加VSMCs培养上清液中CO的生成量,抑制细胞周期进程(P<0.05,P<0.01),并呈时间和剂量依赖趋势;500 mg.L-1的SSTF显著诱导细胞ERK/p-ERK蛋白的表达(P<0.01),但是对HO-1蛋白的表达无影响。结论:SSTF通过阻滞细胞周期进程直接参与抑制VSMCs增殖的作用,ERK信号通路可能参与SSTF介导的细胞保护作用。
OBJECTIVE: To observe the inhibitory effect of SSTF on the proliferation of vascular smooth muscle cells (VSMCs) stimulated by hypertriglyceridemia (HTG) and its possible molecular mechanisms. Methods: Thoracic aorta VSMCs were cultured in vitro and HTG was induced to proliferate. The effects of SSTF on cell proliferation, cell cycle, CO content, HO-1 and ERK / p-ERK protein expression were observed. Results: SSTF at 500 mg.L-1 significantly inhibited the proliferation of VSMCs induced by HTG, increased the production of CO and inhibited the cell cycle progression (P <0.05, P <0.01) in culture supernatant of VSMCs, Dependent manner. SSTF at 500 mg.L-1 significantly induced the expression of ERK / p-ERK protein (P <0.01), but had no effect on HO-1 protein expression. CONCLUSIONS: SSTF is directly involved in the inhibition of VSMCs proliferation by blocking cell cycle progression. ERK signaling pathway may be involved in SSTF-mediated cytoprotection.
