目的:探讨中药一贯煎对肿瘤坏死因子α(TNF-α)致肝炎小鼠模型TNF-α信号通路的干预作用。方法:将C57BL/6小鼠随机分成4组,正常组、模型组、中药组、阳性对照组。正常组及模型组小鼠灌服生理盐水。中药组给予一贯煎汤药灌胃。阳性对照组给予联苯双酯滴丸灌胃。均每天1次,持续1周。模型组、中药组、阳性对照组小鼠腹腔注射0.1μgTNF/12mgD-氨基半乳糖(GaIN)制造肝炎小鼠模型,进行肝功能、肝组织病理、WesternBlot检测TNF-α信号通路相关蛋白cIAP1。结果:一贯煎能减轻肝细胞炎症反应、抑制肝细胞的凋亡、坏死,使小鼠体内ALT和AST水平显著下降(P<0.05);能促进肝细胞cIAP1蛋白的表达(P<0.05)。结论:一贯煎治疗肝炎的药理机制之一在于调节TNF-α信号通路相关蛋白的表达,从而有效减轻炎症反应,达到保肝、降酶的目的。
Objective: To investigate the effect of traditional Chinese medicine on the TNF-α signaling pathway induced by tumor necrosis factor α (TNF-α) in mice induced by hepatitis B virus. Methods: C57BL / 6 mice were randomly divided into 4 groups, normal group, model group, TCM group and positive control group. Normal group and model group mice fed with saline. Chinese medicine group given consistent decoction gavage. The positive control group was given bifendate drip pills. 1 times a day for 1 week. The model mice, the traditional Chinese medicine group and the positive control mice were intraperitoneally injected with 0.1 μg of TNF / 12 mg D-galactosamine (GaIN) to induce hepatitis mouse model. The liver function, liver pathology and Western blot were used to detect the expression of TNF-α signaling pathway related protein cIAP1. Results: The decoction could reduce the inflammatory reaction of hepatocytes, inhibit the apoptosis and necrosis of hepatocytes, significantly reduce the level of ALT and AST in mice (P <0.05), and promote the expression of cIAP1 in hepatocytes (P <0.05). CONCLUSION: One of the pharmacological mechanisms that consistently treat hepatitis is to regulate the expression of TNF-α signaling pathway-related proteins, thereby effectively reducing the inflammatory reaction and achieving the purpose of protecting the liver and lowering the enzyme.
