目的观察熊果酸对矽肺大鼠TNF-α含量和胶原合成的调节作用。方法75只Wistar雄性大鼠,SPF级,随机分为对照组、模型组、熊果酸组,每组25只。模型组与熊果酸组大鼠采用非暴露法气管内一次性注入SiO2悬液(250 mg/kg)复制动物矽肺模型。熊果酸组自注射SiO2第二天灌胃熊果酸40mg/kg,对照组以同样方法灌注等量生理盐水,共连续28天。动物分别在注射SiO2后第7、14、28天处死。ELISA法检测大鼠血清中TNF-α的含量,应用羟脯氨酸试剂盒检测肺组织中总胶原蛋白含量,Western blot法检测Ⅰ/Ⅲ型胶原表达情况。结果与对照组比较,模型组大鼠血清中的TNF-α含量各时间点明显升高,差异有统计学意义(P<0.05)。熊果酸组TNF-α含量与模型组相比均显著降低(P<0.05)。模型组肺组织中总胶原蛋白含量与同期对照组比较明显增加(P<0.05)。熊果酸组与模型组相比,总胶原含量明显下降(P<0.05)。熊果酸组与模型组大鼠肺组织中Ⅰ/Ⅲ型胶原表达与总胶原蛋白含量变化一致。结论熊果酸可以减少矽肺纤维化过程中TNF-α和胶原蛋白的产生。
Objective To observe the regulation of ursolic acid on TNF-α and collagen synthesis in silicotic rats. Methods Seventy - five Wistar male rats were randomly divided into control group, model group and ursolic acid group, 25 rats in each group. Rats in model group and ursolic acid group were injected with SiO2 suspension (250 mg / kg) intratracheally by intratracheal intratracheal injection to establish animal model of silicosis. Ursolic acid group since the injection of SiO2 the next day ursolic acid 40mg / kg, the control group by the same method of perfusion equivalent saline, a total of 28 days. The animals were sacrificed on the 7th, 14th and 28th days after injection of SiO2 respectively. The content of TNF-α in serum was detected by ELISA. The content of total collagen in lung tissue was detected by hydroxyproline kit. The expression of type Ⅰ / Ⅲ collagen was detected by Western blot. Results Compared with the control group, the levels of TNF-α in the serum of the model group were significantly increased at different time points, with significant difference (P <0.05). The content of TNF-α in ursolic acid group was significantly lower than that in model group (P <0.05). The content of total collagen in the lung tissue of the model group increased significantly compared with the control group (P <0.05). Compared with the model group, the total collagen content of ursolic acid group decreased significantly (P <0.05). The expression of type Ⅰ / Ⅲ collagen in the ursolic acid group and model group rats was consistent with that of the total collagen. Conclusion Ursolic acid can reduce the production of TNF-α and collagen in silicosis and fibrosis.
