本试验以亚麻幼苗的下胚轴为外植体进行了卡那霉素选择压力的试验。确定5 0mg/L为适宜的选择压力。同时采用农杆菌介导法 ,利用EHA1 0 5菌株进行了亚麻转基因适宜培养基筛选试验。经试验认为MS培养基是亚麻转基因的适宜培养基。在附加IAA3.5mg/L、KT2mg/L、LH1 5 0mg/L的MS选择培养基上转化外植体的愈伤组织的诱导率可达 77.4%。
In this experiment, the hypocotyls of flax seedlings were used as explants to test the selection pressure of kanamycin. To determine the appropriate selection pressure of 50mg / L. Agrobacterium-mediated method and EHA1057 strain were also used to select appropriate medium for flax transgene. The experiment found that MS medium is a suitable medium for flax transgenics. The callus induction rate of transformed explants on MS selective medium supplemented with IAA3.5mg / L, KT2mg / L and LH150mg / L reached 77.4%.
