目的:探讨间隙连接基因Cx32mRNA、Cx43mRNA及其蛋白产物在肝细胞癌(hepatocelularcarcinoma,HCC)发生的重要意义。方法:应用S-P免疫组织化学法、原位杂交和流式细胞仪分析技术,研究61例HCC、14例正常肝组织以及肝癌细胞系HHCC、SMMC-7721、正常肝细胞系QZG中,Cx32mRNA、Cx43mRNA及其蛋白产物的表达规律。结果:Cx32蛋白在HCCⅠ、Ⅱ、Ⅲ级和正常肝组织中的阳性率分别为55.6%、42.1%、18.2%和92.9%;Cx32蛋白在HHCC、SMMC-7721和QZG中阳性细胞计数率分别为1.9%、1.7%和99.0%。Cx43蛋白在HCCⅠ、Ⅱ、Ⅲ级和正常肝组织中的阳性率分别为44.4%、26.3%、12.1%和78.6%。Cx43蛋白在HHCC、SMMC-7721和QZG中阳性细胞计数率分别为7.3%、2.3%和99.1%。Cx32、Cx43在HCC各级和正常肝组织中差异有显著性(P<0.01),在肝癌细胞系HHCC、SMMC-7721和正常肝细胞系QZG中阳性细胞计数率差异有显著性(P<0.01)。原位杂交显示,Cx32mRNA在HCCⅠ、?
Objective: To investigate the significance of gap junction gene Cx32 mRNA and Cx43 mRNA and their protein products in hepatocellular carcinoma (HCC). Methods: S-P immunohistochemistry, in situ hybridization and flow cytometry were used to investigate the expression of Cx32 mRNA in 61 HCC patients, 14 normal liver tissues and the hepatocellular carcinoma cell lines HHCC, SMMC-7721 and normal liver cell line QZG , Cx43 mRNA and its protein product expression pattern. Results: The positive rates of Cx32 protein in HCC I, II, III and normal liver tissues were 55.6%, 42.1%, 18.2% and 92.9%, respectively. The positive rates of Cx32 protein in HHCC, SMMC-7721 and The positive cell counts in QZG were 1.9%, 1.7% and 99.0%, respectively. The positive rates of Cx43 protein were 44.4%, 26.3%, 12.1% and 78.6% in HCC grade Ⅰ, Ⅱ, Ⅲ and normal liver tissues respectively.
