目的:研究温胆汤(由半夏、茯苓、竹茹、枳实、陈皮、炙甘草配伍)对精神分裂症模型鼠血清超氧化物歧化酶(SOD)、丙二醛(MDA)、一氧化氮(NO)及蛋白激酶C(PKC)含量的影响。方法:将50只清洁级SD大鼠随机分为5组:正常组(A)、模型组(B)、温胆汤高剂量组(C)、温胆汤中剂量组(D)、温胆汤低剂量组(E),每组10只。在造模前,C,D,E组连续ig温胆汤,每次给药20 mL.kg-1(C,D,E组剂量相当于生药40,20,10 g.kg-1);A、B组用等量生理盐水ig,1次/d,共21 d。在最后1次给药2 h后,B,C,D,E组一次性左侧腹腔ip MK801 0.6 mg.kg-1,建立精神分裂症模型;一般状况观察3 d后,处死大鼠,取血清检测。采用生化方法测定血清SOD,MDA和NO含量的改变,用双抗体夹心-酶联免疫吸附(ABC-ELISA)法检测血清中PKC的含量。结果:与模型组比较,温胆汤各组均不同程度改善了模型鼠的一般状况,并且明显降低血清MDA,NO的含量(P<0.05或P<0.01),升高SOD的活性(P<0.05或P<0.01),减轻了氧自由基引起的病理损伤。温胆汤各组显著降低血清PKC的含量(P<0.05或P<0.01)。结论:温胆汤可减轻精神分裂症模型鼠的病理损害,对氧自由基引起的损伤具有一定的保护;降低PKC浓度,从而对缝隙连接通讯(GJIC)的功能起到调节作用。
Objective: To study the effects of Wen Dan Tang (combined with Banxia, Poria cocos, Zhuru, Zhishi, Citrus sinensis and Zhigancao) on serum superoxide dismutase (SOD), malondialdehyde (MDA) Nitrogen (NO) and Protein Kinase C (PKC) Contents. Methods: Fifty clean SD rats were randomly divided into five groups: normal group (A), model group (B), high-dose group of Wendan decoction (C), medium dose group of Wendan decoction (D) Low-dose soup group (E), 10 per group. The content of PKC in serum was detected by double antibody sandwich enzyme-linked immunosorbent assay (ABC-ELISA). Results: Compared with the model group, all the groups of Wendan Decoction improved the general condition of the model rats to varying degrees, and significantly reduced the content of serum MDA and NO (P <0.05 or P <0.01) and increased the activity of SOD (P < 0.05 or P <0.01), reduce the pathological damage caused by oxygen free radicals. Wendan Decoction significantly reduced the content of serum PKC (P <0.05 or P <0.01). Conclusion: Wendan Decoction can alleviate the pathological damage of schizophrenia model rats and protect the injury induced by oxygen free radicals. It can also reduce the concentration of PKC and regulate the function of GJIC.
