青果水提物抑制α-葡萄糖苷酶及改善糖脂代谢作用研究

目的:研究青果水提物对α-葡萄糖苷酶的抑制作用及对糖尿病大鼠血糖、血脂的影响。方法:采用体外酶学的方法评价其对α-葡萄糖苷酶的抑制作用;采用体外细胞培养评价其细胞毒性;采用链脲佐菌素(STZ)结合高脂饲料喂养的方法建立SD大鼠糖尿病模型,将SD大鼠随机分为正常对照组、糖尿病模型组、盐酸二甲双胍阳性对照组及青果水提物高、低剂量组,连续灌胃给药28 d,观察实验大鼠体质量、空腹血糖含量(FBG)、口服葡萄糖耐量(OTGG)及总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)的变化。结果:青果水提物能显著抑制α-葡萄糖苷酶活性(IC50=135.2μg/m L);增强肝细胞活力;缓解糖尿病大鼠体质量减轻,改善口服葡萄糖耐量,降低FBG、TG水平,提高HDL-C水平。结论:青果水提物能显著抑制α-葡萄糖苷酶活性并能有效改善糖尿病大鼠的糖脂代谢。 Objective: To study the inhibition of α-glucosidase by aqueous extract from Fructus Lycii and its effect on blood glucose and blood lipid in diabetic rats. Methods: The inhibitory effect on α-glucosidase was evaluated by in vitro enzymatic method. The cytotoxicity of α-glucosidase was evaluated by in vitro cell culture. SD rats were established by streptozotocin (STZ) combined with high-fat diet. Model, the SD rats were randomly divided into normal control group, diabetic model group, metformin hydrochloride positive control group and high fructose aqueous extract group, low dose group, continuous gavage for 28 days, the body weight, fasting blood glucose FBG, OTGG, TC, TG, HDL-C and LDL-C were measured. Results: The aqueous extract of Fructus Mume could significantly inhibit the activity of α-glucosidase (IC50 = 135.2μg / m L), enhance the viability of hepatocytes, alleviate the weight loss of diabetic rats, improve the oral glucose tolerance, decrease the levels of FBG and TG HDL-C levels. Conclusion: The fruit extract can significantly inhibit the α-glucosidase activity and can effectively improve the diabetic rats glucose and lipid metabolism.
